Summary of the Study

Accurate characterization of the tumor microenvironment (TME), including immune cell distribution and interactions, is crucial for improving immunotherapy outcomes. This study introduces a spatial analytics tool built around Brightplex® multiplex IHC, digital pathology, and custom R-based analytics.

Key contributions:

  • Developed spatial analysis features using the spatstat R package, including:

    • Proximity metrics
    • G-function interaction scores
    • Kernel density (heatmap)–based spatial mapping
  • Integrated spatial analysis across serial slides stained with distinct Brightplex® multiplex panels

  • Demonstrated proximity of Tregs to CD8+ T cells and macrophages to tumor cells as potential predictors of outcome

  • Designed to support clinical decision-making and biomarker discovery through integration with the Immunogram framework

Important

In this study, Alboukadel Kassambara contributed to the analysis and software development, building a dedicated R package for spatial analytics. He implemented the bioinformatics workflow, standardized the data pipeline, and integrated it with Brightplex® multiplex staining to enable robust, reproducible spatial quantification of tumor–immune cell interactions.
